SyBr Green analysis of parasite lifecycle. The culture was initiated by incubating mature schizonts with RBCs in a complete medium at 1% haematocrit. Aliquots were taken at the indicated time points (0- to 94-hr) and centrifuged on 40% Percoll cushions to remove haemozoin and cell debris. Cell pellets were then used in the SyBr Green I assay either with or without RNase A treatment to determine the effects of RNA transcripts on RFI readings. Three biological replicates were run in triplicate and averaged, and the standard deviations calculated. Please see Additional file 5 for photos of lifecycle stages.
