Each reaction was performed in 2 independent experiments in a standard reaction buffer containing 20 mM Tris–HCl, pH 8.0, 1 mM DTT, 1 mM EDTA, 50 mM NaCl, 100 μg/ml BSA, 2.5 nM recombinant PfUDG, and 0.01-3.5 μM ³²P-labelled single strand oligo-U (ss-U) or double stranded A:U (ds-A:U) at 37°C for 10 min.
