PfWLP1 is intracellularly located in gametocytes. a Detection of plasma membrane-bound proteins. Gametocytes were either stripped from the enveloping erythrocyte and parasitophorous vacuole membranes by saponin-treatment prior to fixation or were membrane-permeabilized following fixation. The cells were immunolabelled with antibodies directed against PfWLP1, Pfs230, PfGAP50 or the HA-tag (green); the gametocytes were counterstained with Evans Blue (red). The parasite nuclei were highlighted by Hoechst nuclear stain (blue). Bar 5 µm. b Quantification of immunolabelling. A total number of 50 gametocytes were evaluated for immunolabelling as described in a in triplicate. Data are representative of three independent experiments
