Schematic overview of function and localisation of Ang-1, Ang-2 and the Tie-2 receptor. Ang-2 is pre-stored in the Weibel–Palade bodies in endothelial cells and is released upon endothelial cell activation. Ang-2 replaces Ang-1 by binding the Tie-2 receptor, preventing its activation and thereby blocking the anti-inflammatory, anti-apoptotic and tight-junction supporting effects of Ang-1.
