Mid- and long-term effects of ML304 on the glutathione redox ratio of Plasmodium falciparum NF54[hGrx1–roGFP2]. 4 h incubation of NF54[hGrx1–roGFP2] -transfectants with ML304 led to a concentration-dependent increase in fluorescence ratio as detected using the Clariostar plate reader (a). 24 h incubation with varying ML304 concentrations led to increases in 405/475 nm fluorescence ratios with a significant effect of the highest compound concentration (8.88 µM) (b). Non-treated parasites served as controls. All experiments included fully oxidized (1 mM DIA) and fully reduced (10 mM DTT) parasites (each using 2 min incubations) prior to blocking with NEM. Mean values and standard errors of the means (± SEM) are shown for three independent experiments. A one-way ANOVA test with 95% confidence intervals with Dunnett’s multiple comparison test was applied for statistical analysis of significance (**p < 0.01; ***p < 0.001)
