(A and B) show the peaks of thiosemicarbazide, free PLP, and PLP mixed with thiosemicarbazide (PLP-thiosemicarbazone) at the retention times 0.89, 1.28 and 3.56 min, respectively. (C and D) show the peaks of the filtrate obtained from a mixture of PvSHMT (62.79 μM) with thiosemicarbazide (10 mM), which was mixed with SDS (1% w/v) and separated by a Centricon filtration unit (10 kDa cut-off). A peak with a retention time of 3.56 min was observed.
