Enrichment of P. falciparum O-GlcNAcylated proteins was performed by utilizing sWGA in conjunction with PNGase F digestion (a). As a second approach, after labelling of P. falciparum O-GlcNAcylated proteins by Click-chemistry as described previously, clicked proteins were treated or not with PNGase F, purified and enriched using avidin beads for further identification by mass spectrometry (b). Gels were brilliant blue-stained. Bands were excised, submitted to trypsin digestion for further identification
